Els medicaments disponibles actualment nom\u00e9s poden ajudar a controlar els s\u00edmptomes de la malaltia, per\u00f2 no poden prevenir el deteriorament f\u00edsic, mental i conductual associat amb l&#8217;afecci\u00f3.<br><br>Despr\u00e9s de mostrar l&#8217;impacte de la desregulaci\u00f3 de la senyalitzaci\u00f3 entre neurones i micr\u00f2glia a trav\u00e9s <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/31790427\/\">d&#8217;alteracions en el patr\u00f3 d&#8217;expressi\u00f3 temporal de CD200-CD200R1<\/a>, els cient\u00edfics de Creatio han descobert la possible implicaci\u00f3 d&#8217;un altre eix molecular que uneix el sistema immune amb la HD. Els resultats acaben de <a href=\"https:\/\/www.frontiersin.org\/articles\/10.3389\/fncel.2020.00163\/full\">publicar-se en Frontiers in Cellular Neuroscience<\/a>. En el cervell sa, la micr\u00f2glia, les c\u00e8l\u00b7lules immunes residents del sistema nervi\u00f3s central, es troba en un estat de vigil\u00e0ncia constant, escanejant cont\u00ednuament l&#8217;entorn cerebral. Aquest estat de vigil\u00e0ncia es mant\u00e9 a trav\u00e9s de la interacci\u00f3 entre el lligant fraktalina (FKN), alliberat per neurones i el receptor CX3CR1 expressat en micr\u00f2glia. En el cervell dels pacients amb HD, es coneix que la micr\u00f2glia s&#8217;activa anys abans que apareguin les manifestacions cl\u00edniques, tot i que la naturalesa d&#8217;aquesta activaci\u00f3 i el possible impacte benefici\u00f3s o perjudicial encara \u00e9s dif\u00edcil d&#8217;assolir. A l&#8217;article, Creatio mostra que els nivells de FKN s\u00f3n regulats a la baixa en el putamen de pacients amb HD, a les c\u00e8l\u00b7lules mare derivades de pacients i en l&#8217;estriat dels models de ratolins de la malaltia, el R6 \/ 1. A m\u00e9s, l&#8217;an\u00e0lisi automatitzat de la morfologia cel\u00b7lular utilitzant microsc\u00f2pia confocal de dos fotons i el programari 3DMorph va permetre mostrar una morfologia microglial ramificada no inflamat\u00f2ria en l\u2019estriat dels ratolins R6 \/ 1. Aquests resultats apunten cap a un augment anormal en l&#8217;embolcall sin\u00e0ptica microglial en animals pre-simptom\u00e0tics per HD que podria contribuir a la p\u00e8rdua progressiva de sinapsis i disfunci\u00f3 estriatal. A m\u00e9s, l&#8217;an\u00e0lisi morfol\u00f2gic es va complementar amb registres electrofisiol\u00f2gics que van demostrar que la manca de FKN est\u00e0 involucrada en la disfunci\u00f3 de plasticitat sin\u00e0ptica estriatal en ratolins pre-simptom\u00e0tics R6 \/ 1. Tenint en compte tot en el seu conjunt, els resultats publicats per Creatio i col\u00b7laboradors revelen un paper clau per a la senyalitzaci\u00f3 de FKN-CX3CR1 en la fisiopatologia de la HD: els d\u00e8ficits de senyalitzaci\u00f3 de FKN-CX3CR1 a la HD condueixen a interaccions anormals neurona-micr\u00f2glia, que poden contribuir a la disfunci\u00f3 de plasticitat sin\u00e0ptica estriatal primerenca, caracter\u00edstica d&#8217;aquesta malaltia neurol\u00f2gica. Curiosament, s&#8217;ha descrit un major embolcall de material sin\u00e0ptic intervinguda per micr\u00f2glia en models de ratol\u00ed d&#8217;altres malalties neurol\u00f2giques com l&#8217;Alzheimer, l&#8217;esquizofr\u00e8nia i el s\u00edndrome de Rett.
