Comella-Bolla, han realitzat la caracteritzaci\u00f3 en profunditat d&#8217;un protocol de diferenciaci\u00f3 <em>in vitro<\/em> que produeix neurones funcionals del cervell anterior, incloent-hi neurones de projecci\u00f3 estriatals (MSNs, de l&#8217;angl\u00e8s medium spiny neurons) a partir de c\u00e8l\u00b7lules mare pluripotents humanes (hPSCs) en nom\u00e9s 37 dies. <\/h3>\n\n\n\n<p>En l&#8217;article publicat ara a <em>Molecular Neurobiology<\/em>, Comella i col\u00b7laboradors descriuen amb precisi\u00f3 les diferents fases de diferenciaci\u00f3, aix\u00ed com les mol\u00e8cules utilitzades per a induir aquesta diferenciaci\u00f3, mitjan\u00e7ant el control de la inducci\u00f3 neural, la regionalitzaci\u00f3, la neurog\u00e8nesi i la sinaptog\u00e8nesi. El proc\u00e9s de diferenciaci\u00f3 s\u2019ha seguit i caracteritzat mitjan\u00e7ant t\u00e8cniques de rastreig cel\u00b7lular en massa (<em>high-cell content<\/em>), t\u00e8cniques g\u00e8niques d&#8217;alt rendiment quantitatiu (<em>high-throughput<\/em>) com la plataforma d\u2019<a href=\"https:\/\/www.cell.com\/molecular-therapy-family\/methods\/fulltext\/S2329-0501(16)30042-0?_returnURL=https%3A%2F%2Flinkinghub.elsevier.com%2Fretrieve%2Fpii%2FS2329050116300420%3Fshowall%3Dtrue\" target=\"_blank\" rel=\"noreferrer noopener\">OpenArray<\/a> i d\u2019imatge funcional mitjan\u00e7ant els corrents de calci (<em>high-throughput calcium imaging<\/em>). El protocol de Creatio mostra la reproducci\u00f3 dels canvis din\u00e0mics en l&#8217;expressi\u00f3 g\u00e8nica que tenen lloc durant el desenvolupament neurol\u00f2gic hum\u00e0. A m\u00e9s, els fenotips funcionals de les neurones obtingudes es confirmen mitjan\u00e7ant l&#8217;expressi\u00f3 de marcadors sin\u00e0ptics, canals i\u00f2nics activats per voltatge i receptors de neurotransmissors mentre es mostren diversos patrons d&#8217;activitat espont\u00e0nia.<\/p>\n\n\n\n<p>Considerant totes aquestes caracter\u00edstiques, estem conven\u00e7uts que aquest protocol \u00e9s una plataforma ideal per a l&#8217;estudi <em>in vitro<\/em> tant del neurodesenvolupament com de les malalties neuronals mitjan\u00e7ant la nova <a href=\"http:\/\/www.avantdrug.eu\/\" target=\"_blank\" rel=\"noreferrer noopener\">plataforma AVANTDRUG\u00ae<\/a>.\u00a0<\/p>\n\n\n\n<p>\u00c9s important destacar que el protocol de Creatio s&#8217;ha desenvolupat i validat seguint les estrictes pautes establertes en la ISO9001 i per l&#8217;<a href=\"https:\/\/www.oecd.org\/\" target=\"_blank\" rel=\"noreferrer noopener\">OCDE<\/a> en el <em>Guidance Document on Good In Vitro Method Practices<\/em> (<a href=\"https:\/\/www.oecd.org\/env\/guidance-document-on-good-in-vitro-method-practices-givimp-9789264304796-en.htm\" target=\"_blank\" rel=\"noreferrer noopener\">GIVIMP<\/a>).
