L\u2019equip de Creatio demostra com la implantaci\u00f3 d\u2019un sistema de gesti\u00f3 de qualitat (SGQ) promou l&#8217;estabilitzaci\u00f3 gen\u00f2mica a trav\u00e9s de les l\u00ednies de c\u00e8l\u00b7lules mare pluripotents humanes (hPSC), produint resultats cient\u00edfics d&#8217;alta qualitat i reprodu\u00efbles.<\/h3>\n\n\n\n<p>Actualment, la recerca amb c\u00e8l\u00b7lules mare \u00e9s molt prometedora i compta amb un gran nombre d\u2019aplicacions, des del descobriment de nous f\u00e0rmacs, passant per la medicina regenerativa, models de malalties fins a l\u2019estudi de mecanismes de desenvolupament embrionari, entre altres.&nbsp; A causa d\u2019aquest nombre d\u2019aplicacions, els articles que utilitzen hPSC, els quals inclouen tant les hESC com les hiPSC, s&#8217;han multiplicat per 56 en els darrers 30 anys. Malauradament, aquest creixement no s&#8217;ha relacionat amb l&#8217;estandarditzaci\u00f3 dels protocols in vitro utilitzats. Unes condicions de cultiu sub\u00f2ptimes limiten la reproductibilitat dels resultats, a m\u00e9s d\u2019estar relacionades amb alteracions gen\u00f2miques que poden posar en perill la translaci\u00f3 cl\u00ednica. Per exemple, la detecci\u00f3 de noves alteracions va provocar la finalitzaci\u00f3 de l&#8217;assaig cl\u00ednic RIKEN el 2015. RIKEN \u00e9s el primer estudi que utilitzava c\u00e8l\u00b7lules hiPSC per tractar pacients amb degeneraci\u00f3 macular relacionada amb l&#8217;edat (DMAE).&nbsp;<\/p>\n\n\n\n<p>La comunitat cient\u00edfica n\u2019\u00e9s conscient, malgrat que nom\u00e9s 1 de cada 20 articles analitzats descriu la integritat gen\u00f2mica de les hPSC utilitzades. En aquest article, per tal d\u2019entendre amb quina freq\u00fc\u00e8ncia apareixen aquestes alteracions i estudiar les seves implicacions, els investigadors de Creatio van utilitzar resultats d&#8217;estudis existents, i els van complementar amb dades pr\u00f2pies obtingudes durant 5 anys (2017 \u2013 2021). A m\u00e9s, van investigar si la implementaci\u00f3 de la ISO9001:2015 pot millorar la qualitat de les dades reduint la quantitat d\u2019alteracions gen\u00f2miques en les hPSC. Per fer-ho, van comparar una hibridaci\u00f3 gen\u00f2mica comparativa basada en matrius (aCGH en angl\u00e8s, amb una resoluci\u00f3 d&#8217;1 kb) amb una t\u00e8cnica m\u00e9s assequible per\u00f2 menys sensible (G-banding, que detecta canvis superiors a 5 Mb), els investigadors van detectar alteracions en 8 hESC i 11 l\u00ednies hiPSC , abans i despr\u00e9s de l&#8217;adaptaci\u00f3 al SGQ.&nbsp;<\/p>\n\n\n\n<p>Els resultats observats i publicats a <a href=\"https:\/\/www.mdpi.com\/2073-4409\/11\/13\/1984\/review_report\" target=\"_blank\" rel=\"noreferrer noopener\">Cells<\/a>&nbsp;van mostrar que, tot i que, les hiPSC adquirien m\u00e9s mutacions (2 vegades m\u00e9s altes pel pas 42 que les hESC), aquestes s\u00f3n majorit\u00e0riament mutacions no pat\u00f2genes en comparaci\u00f3 amb les observades a les hESC.&nbsp; Al llarg del temps, les hiPSC es van mantenir m\u00e9s estables que les hESC, fet que les converteix en millors candidates per a futures aplicacions tant cl\u00edniques com de recerca. Curiosament, despr\u00e9s d\u2019implementar el SGQ, la qualitat de les l\u00ednies cel\u00b7lulars va millorar significativament. Per a les c\u00e8l\u00b7lules hiPSCs del pas 7, l&#8217;adquisici\u00f3 d&#8217;alteracions gen\u00f2miques potencialment pat\u00f2genes es va reduir 4 vegades. Els resultats mostren la necessitat d&#8217;implementar un SGQ quan es treballa amb hPSC, especialment en camps on es requereixen m\u00e8todes humans alternatius com s\u00f3n les neuroci\u00e8ncies, on els models animals no han demostrat equival\u00e8ncia amb humans.&nbsp;<\/p>\n\n\n\n<p>Francisco Molina, autor principal de l&#8217;article publicat a Cells, assenyala que \u00abaquest treball ofereix una visi\u00f3 integral de les acumulacions d&#8217;alteracions durant el cultiu prolongat de hPSC i, per primera vegada, d\u00f3na suport a la necessitat d&#8217;estandarditzar les pr\u00e0ctiques de cultiu in vitro\u00bb. El candidat <a rel=\"noreferrer noopener\" href=\"https:\/\/www.asctn-training.net\/\" target=\"_blank\">Marie-Curie ASCTN-ITN<\/a> Early Stage Researcher (ESR) espera que la comunitat cient\u00edfica valori la import\u00e0ncia d&#8217;implementar SGQ als laboratoris que realitzen recerca amb hPSC.&nbsp;<\/p>\n\n\n\n<p>Creatio, la instal\u00b7laci\u00f3 de producci\u00f3 de medicina regenerativa, t\u00e9 una forta cultura translacional.
