This is the body\u2019s first line of defence, and macrophages \u2014 cells of the innate immune system \u2014 play a key role: first, they help eliminate pathogens and other infectious agents, and then they trigger the mechanisms that repair the damage caused during the inflammatory process.<\/strong><\/p>\n\n\n\n<p>Now, a study by the University of Barcelona, the findings of which have been published in the journal <a href=\"https:\/\/www.cell.com\/cell-reports\/fulltext\/S2211-1247(26)00159-2?_returnURL=https%3A%2F%2Flinkinghub.elsevier.com%2Fretrieve%2Fpii%2FS2211124726001592%3Fshowall%3Dtrue\"><em>Cell Reports<\/em><\/a>, has discovered that a protein called Pol\u03bc is essential to this repair process, as it enables macrophages to survive at the site of inflammation. According to the researchers, a deficiency of this protein could be the cause of certain autoinflammatory diseases, conditions in which the immune system is activated inappropriately, leading to chronic inflammation and tissue damage.<\/p>\n\n\n\n<figure class=\"wp-block-image\"><img decoding=\"async\" src=\"https:\/\/web.ub.edu\/documents\/2710030\/46107403\/macrofags_destacada.jpg\/cfa44ca9-7367-1e07-5c9c-79b031fc5f6f?t=1777020659385\" alt=\"From left to right, researchers Carlos Sebasti\u00e1n, Jorge Lloberas, Carlos Batlle and Antonio Celada.\"\/><\/figure>\n\n\n\n<p>From left to right, researchers Carlos Sebasti\u00e1n, Jorge Lloberas, Carlos Batlle and Antonio Celada.<\/p>\n\n\n\n<p>The study, conducted using animal models, was led by researchers Jorge Lloberas, Antonio Celada and Carlos Sebasti\u00e1n, all three members of the Immunology Section of the Department of Cell Biology, Physiology and Immunology at the Faculty of Biology, the Barcelona Science Park-UB (PCB-UB) and the Consolidated Research Group on Inflammation: Basic and Translational Research (InFlam-BaTra). Carlos Sebasti\u00e1n is also a member of the UB Institute of Biomedicine (IBUB) and is currently the principal investigator of the Metabolic Dynamics in Cancer Group.<\/p>\n\n\n\n<p>Carlos Batlle-Recoder is the lead author of the study, which included the participation of the National Centre for Biotechnology (CNB-CSIC).<\/p>\n\n\n\n<p><strong>The collateral damage caused by defence mechanisms<\/strong><\/p>\n\n\n\n<p>When the innate immune system triggers the inflammatory response to an external threat, macrophages engulf the pathogens and produce large quantities of reactive oxygen species (ROS) to destroy them. However, ROS also have a harmful effect on the macrophages themselves, as they can induce DNA damage that may lead to the death of these defence cells and promote chronic inflammation.<\/p>\n\n\n\n<p>Previous studies had demonstrated the importance of Pol\u03bc in lymphocyte development and its involvement in the repair of genetic damage, but until now, its role in macrophages and in the inflammatory response was unknown. The researchers have discovered this by analysing the protein\u2019s role in this process using animal models of skin inflammation and muscle injury. \u201cUsing mice lacking Pol\u03bc, we have been able to demonstrate, in both in vitro and in vivo experiments, that the two phases of the inflammatory response are defective in the absence of this polymerase,\u201d they explain.<\/p>\n\n\n\n<p>The study shows, in animal models, that a deficiency of this protein compromises the survival of essential immune system cells.<\/p>\n\n\n\n<p><strong>Implications for disease and potential therapeutic strategies<\/strong><\/p>\n\n\n\n<p>In light of these findings, the researchers suggest that a deficiency in the Pol\u03bc protein could underlie certain diseases, such as interferonopathies, which are characterized by autoinflammatory processes that chronically activate type I interferons \u2014 key molecules of the immune system that coordinate the response to viral infections \u2014 causing damage to tissues and organs. Although no inflammatory conditions associated with Pol\u03bc have been described in humans to date, the researchers suggest that this may be because this possibility has not yet been sufficiently studied in certain clinical contexts. \u201cIt is possible that, in the case of some inflammatory conditions, the presence of mutations in Pol\u03bc has simply not been analysed,\u201d they note.<\/p>\n\n\n\n<p>Furthermore, the findings could also have implications in other situations, such as cases of macrophage hyperactivity, as occurs during septic shock. \u201cIn such cases, Pol\u03bc activity could be inhibited to reduce the excessive activity of these cells, which could help to lower patient mortality,\u201d the researchers note.
