D<\/strong><strong>\u00a0(VDRs)\u00a0<\/strong>median probablemente todas las acciones de la vitamina D en vertebrados superiores. El calcitriol, sintetizado en el ri\u00f1\u00f3n y varios tipos de c\u00e9lulas epiteliales y del sistema inmune, es en realidad una hormona pleiotr\u00f3pica con numerosas acciones reguladoras en el organismo. Entre ellas, la regulaci\u00f3n de los niveles de calcio y fosfato, y la biolog\u00eda y mineralizaci\u00f3n \u00f3sea.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-37f3c25 elementor-widget elementor-widget-heading\" data-id=\"37f3c25\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Clasificaci\u00f3n<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-c598de6 elementor-widget elementor-widget-text-editor\" data-id=\"c598de6\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Los VDRs son miembro de la\u00a0<strong>subfamilia NR1<\/strong>, de la gran superfamilia de\u00a0receptores nucleares\u00a0(NRs), junto con los receptores\u00a0<a href=\"https:\/\/www.ub.edu\/nurcamein\/es\/receptores-nucleares\/tr-receptor-de-hormona-tiroidea\/\">TRs<\/a>\u00a0y RARs.<\/p><p>A su vez, en base a su afinidad por el ligando y a su modo de acci\u00f3n,\u00a0est\u00e1n clasificados\u00a0como\u00a0<strong>receptores endocrinos de Clase II<\/strong>, formando heterod\u00edmeros con\u00a0<a href=\"https:\/\/www.ub.edu\/nurcamein\/es\/receptores-nucleares\/rxr-receptor-x-retinoide\/\">RXR<\/a>.\u00a0<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-c6d67e5 elementor-widget elementor-widget-heading\" data-id=\"c6d67e5\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Estructura e interacciones<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-e64f411 elementor-widget elementor-widget-text-editor\" data-id=\"e64f411\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Todos los miembros de la superfamilia de receptores nucleares comparten una\u00a0estructura general\u00a0multidominio. Los VDRs tienen los siguientes dominios:<\/p><ul><li><strong>Dominio A\/B o\u00a0<i>Activation Function-1<\/i>\u00a0(NTD\/AF-1):<\/strong>\u00a0<b>regi\u00f3n de uni\u00f3n a coactivadores en posici\u00f3n N-terminal, con funci\u00f3n de transactivaci\u00f3n g\u00e9nica dependiente o independiente del ligando.<\/b><\/li><li><strong>Dominio C (DBD):<\/strong>\u00a0regi\u00f3n de uni\u00f3n al DNA altamente conservada,\u00a0que contiene dos \u00abdedos de zinc\u00bb que interaccionan con secuencias de nucle\u00f3tidos espec\u00edficas en los genes diana denominados elementos de respuesta hormonal (motivos HREs, en este caso denominados\u00a0<i>Vitamin D Response Elements\u00a0<\/i>o VDREs). El heterod\u00edmero VDR-RXR se une a los elementos de respuesta a vitamina D (VDRE) ubicados en el promotor, regi\u00f3n codificante e intrones, o a veces muy alejados de los genes diana, regulando su\u00a0 velocidad de transcripci\u00f3n.<\/p><p>La uni\u00f3n del ligando a VDR modifica la conformaci\u00f3n de su regi\u00f3n LBD, promoviendo habitualmente la liberaci\u00f3n de corepresores transcripcionales y el reclutamiento de coactivadores y de enzimas (acetilasas de histonas\u2026 ) que modifican la estructura de la cromatina, facilitando la activaci\u00f3n transcripcional de los genes diana. En otros casos el efecto es opuesto, se produce una represi\u00f3n transcripcional por mecanismos no bien conocidos.<\/p><h4>V\u00eda no-gen\u00f3mica<\/h4><p>La uni\u00f3n del ligando a la poblaci\u00f3n citos\u00f3lica de VDR desencadena m\u00faltiples v\u00edas o cascadas de se\u00f1alizaci\u00f3n intracelular, que dan como resultado respuestas inmediatas independientes de la transcripci\u00f3n g\u00e9nica en las c\u00e9lulas.
