Molecular basis of membrane traffic in mammalian cells - PI: Carles Enrich Bastús

In the last years, our research group has focused on fundamental aspects of the traffic of proteins, metabolism and signaling at the cellular level, and the in vivo consequences in animal models. Intracellular compartments and organelles maintain interactions that are important for regulating metabolic and cellular processes. In this sense, it has been shown that the AMPK microtubule-dynamics regulation allows the interaction of mitochondria and lipid droplets to ensure energy requirements when fasting. On the other hand, it has been demonstrated that the AnxA6 located in the endocytic compartment regulates intracellular cholesterol levels and its cellular homeostasis. In situations such as NPC1 disease, or experimentally after overexpressing AnxA6, cholesterol accumulates in endosomes. This induces mislocalization of a group of SNARE proteins, interfering the recycling of integrins, and consequently generating a reduction in cell migration and invasion. We have also analyzed the dynamic’s association of small GTPases, such as KRas and Rac1, with endosome membranes, and the results have highlighted the role of these proteins in the endocytic compartment signaling to regulate cellular processes such as cell migration.

Currently, our research is organized into 3 main interdisciplinary lines to understand the molecular mechanisms that translate cell surface originated signals into metabolic and functional changes within the cells. Our research groups are:

  • Team: Endocytosis and intracellular trafficking of proteins and cholesterol (IP: Carles Enrich)

Our group is interested on the molecular mechanisms of membrane traffic in mammalian cells. This basic research is important for understanding a number of diseases including cancer, diabetes, and heart disease or liver disorders. We study the regulation of a variety of aspects of protein and cholesterol trafficking and signal transduction in the endocytic compartment. We focus on determining the role of annexin A6 in signalling, membrane trafficking and liver regeneration and glucose metabolism.

Lipid droplets are organelles that collect, store and supply lipids in all eukaryotic cells. These organelles are also cellular hallmarks of prevalent diseases ranging from obesity to cancer. The cell biology of lipid droplets and its role in pathogenesis are still unclear. Our group is interested in the mechanisms of lipid droplet biogenesis and consumption. In this sense, the aim of our project is to characterize - in health and in disease - the cellular processes that are regulated or altered due to changes of the lipid fluxes existing in our cells. For instance, cholesterol accumulation promotes mitochondrial dysfunction and oxidative damage underlying in the pathogenesis of steatohepatitis, neurodegeneration and atherosclerosis. In this context, we are especially interested in how caveolin-1 regulates cholesterol trafficking and storage.

 

  • Team: Signalling, cytoskeleton and cellular migration (IP: Francesc Tebar)

Our group is studying the signaling and trafficking of the epidermal growth factor receptor (EGFR) and the small GTPases KRas and Rac1. The signaling elicited by these proteins controls various important cellular processes such as proliferation, cell migration and mobility. EGFR signaling is initiated at the plasma membrane and remains in the endosomes, these signaling platforms can move to different cellular locations, thereby restricting spatial signaling and controlling different processes in the cell. Accordingly, many signaling proteins of EGFR have been detected in endosomes, among them KRas and Rac1. One of the objectives of the group is to study the involvement of the endocytic compartment on EGFR, KRas and Rac1 functionality.
